Fluorophores
A fluorophore is a molecule with Fluorescence properties. The fluorophore absorbs photons and emits photons of lower energy in return. Fluorophores are chemically diverse. Fluorophores relevant in the life sciences range from small chemical compounds (fluorescein, rhodamine, cyanine and their derivatives) to amino acids (tryptophan, tyrosine, phenylalanine) to Fluorescent fusion proteins (GFP, RFP etc.).Chemical fluorophores are often attached to biomolecules via covalent or tag-specific Protein Labeling procedures, while fluorescent proteins can be genetically fused to the target protein.
NanoTemper Technologies offers Protein Labeling kits with different coupling chemistries (amine and cysteine reactive) which are optimized for TRIC experiments. In addition, NanoTemper Technologies exclusively offers highly specific labeling kits for His-tags (RED- tris-NTA). In addition to small chemical labels, fluorescent fusion proteins can be used in TRIC experiments.
Note: When choosing a fluorophore t, ensure it is compatible with the Dianthus Filterset. Excitation and emission wavelengths often vary depending on the environment (buffer, pH). All NanoTemper Technologies RED Protein Labeling kits are compatible with Dianthus. The NanoTemper Technologies RED fluorophores are also available as RED-tris-NTA variants for convenient labeling of a His-tag.
Tip : While 2nd Generation labeling kits provided by NanoTemper generally yield better results, in some instances it may be necessary to test the first generation of NanoTemper Technologies fluorophores, when a binding assay cannot be established with the 2nd Generation kits.
