The group of Kaspar Locher from the ETH Zurich shows that MicroScale Thermophoresis (MST) is a suitable tool for the analysis of interactions between detergent-solubilized membrane proteins and their substrates.
As presented in this application note, they analyzed the interaction of peptides and the membrane enzyme PglB. During the catalytic cycle of enzymatic reactions, the recognition of a specific substrate by the enzyme is one of the most important steps. Thus, the analysis of the affinity between enzyme and substrates as well as between enzyme and inhibitors provide important insights into mechanistic biochemistry of the N-glycosylation reaction catalyzed by the PgIB enzyme.
