Dianthus PicoDuo instruments have the great advantage of having two optical systems that can work in parallel to measure Plates, thereby reducing the average time to read a plate from ~1 h to ~30 min for an On-time of 5 sec per well. Note that there are some Plate Layouts that do not allow the simultaneous use of both optics. The two optical systems are called top and bottom, corresponding to the part of the plate they are responsible for. The below figure shows the orientation of the optics and the typical direction in which Dianthus PicoDuo instruments read assigned Wells.

In the DI.Screening Analysis software the optics used to measure a certain well can be seen in the ligand table. The column with that information is not active by default and must be actively selected under “Choose Columns”.
