Protein concentration and DOL
After Protein Labeling with a fluorescent dye (see Fluorophores ), it is strongly recommended to determine the protein concentration and labeling efficiency. The degree of labeling (DOL) specifies how many dye molecules are attached to one protein molecule in the sample. A DOL of around 1 is recommended. To calculate the DOL, use a UV/VIS spectrophotometer to measure an absorbance spectrum of the sample. The NanoTemper Technologies Labeling Kits come with detailed instructions how to calculate DOL.
